Summary: | The entomopathogenic fungal isolate Paecilomyces fumosoroseus Pfr116 highly infective to sucking insects was cultured in submerged conditions at 25℃ using five liquid media. Based on the yield of dry mycelia over incubation time, a low-cost medium was selected for use in mass production from the five media tested. Its components (w/v) included 2% sucrose, 20% potato, and 0.1% peptone supplemented with 0.01% NaNO<sub>3</sub>, 0.01% KCl, 0.025% MgSO<sub>4</sub>, 0.005% MnSO<sub>4</sub>, 0.005% FeSO<sub>4</sub> and 0.05% KH<sub>2</sub>PO<sub>4</sub>. A 48-h culture of the medium resulted in a mycelial yield of>20 mg/mL, which was close to the mycelial yield of Sabouraud dextrose broth during the same period of incubation. Other optional conditions for initiating the culture included: ① the amount of initial inoculum to guarantee the liquid containing mycelial biomass of 1.5-2.0 mg/mL; and ② a pH ranging from 5 to 7. |
