EGCG ((—) Epigallocatechin gallate), one of the catechins in tea, is the most active natural polyphenol in the products and, being a polyligand, can complex with Cu<sup>2+</sup>. In this paper, the reaction condition for EGCG to complex with Cu<sup>2+</sup> was investigated. The results showed that pH value and the ratio of EGCG to Cu<sup>2+</sup> were the main influencing factors. Under the condition of strong acidity (pH≤4.0), it was difficult for EGCG to complex with Cu<sup>2+</sup>; at 9.0>pH>4.0, EGCG could complex with Cu<sup>2+</sup>; at pH>9.0, catechol-hydroxyl groups interacted with Na<sup>+</sup> to form catechol-Na<sup>+</sup>. Presence of Cu<sup>2+</sup> strongly promoted the autoxidative activity of EGCG, so that Cu<sup>2+</sup> was oxidized to Cu<sup>+</sup>.In the meantime we found that in acetic acid buffer solution of pH6.0 the ratio of Cu<sup>2+</sup> to EGCG in the complex was 2∶1 and in boric acid buffer solution of pH7.4 the ratio was 1∶1. When Cu<sup>2+</sup> to EGCG ratio was above 4∶1. redox reaction occurred resulting in promoting the autoxidative activity of EGCG by Cu<sup>2+</sup>. |
