description WON-21, an herbal formulation used to alleviate hangover symptoms, has beneficial effects on oxidative stress, inflammation, and alcohol-metabolizing enzymes. This study evaluated the protective effects of WON-21 against ethanol-induced gastritis. In vitro, WON-21 and its constituents, quercetin and apigenin, inhibited TNF-α-induced IL-8 production in human gastric adenocarcinoma cells. In vivo, ethanol-induced gastritis was modeled in rats. Oral pretreatment with 200 and 500 mg/kg WON-21 reduced gastric mucosal lesions by 61.8 % and 79.3 %, respectively. Histopathological analysis revealed a dose-dependent protective effect of WON-21 against inflammatory cell infiltration and epithelial damage. WON-21 also significantly decreased IL-6 levels and suppressed the mRNA expression of IL-8 and IL-18 in gastric tissues. Network pharmacology analysis was performed to elucidate key targets and pathways, supporting these findings. The gastroprotective efficacy of the high dose (500 mg/kg) was comparable to that of omeprazole. These results demonstrate that WON-21 exerts anti-inflammatory and mucosa-preserving effects against ethanol-induced gastritis.
